Screening of nutrient parameters for ganoderic acid production by Ganoderma Lucidum Nrcm oe 52 under submerged fermentation using Plackett-Burman design

Ganoderic acid is a triterpenoid, anticancer produced
by Ganoderma lucidum. In the present research
the Plackett- Burman experimental design was used to
identify the important medium constituent influencing
the production of ganoderic acid B by Ganoderma
lucidum NRCM OE 52 under submerged fermentation.
Nine medium parameters such as maltose, glucose, corn
flour, MnSO4.H2O, urea, soya flour, NaCl, biotin and
Penicillium citrinum extract as elicitor were screened by
Plackett ? Burman Design in 12 experimental run which
include 9 medium parameters and 2 dummy variables.
Nine nutrient parameter were maltose, glucose, corn
flour, MnSO4.H2O, urea, soya flour, NaCl, biotin and
elicitor shows 1.80 %, 0.96 %, 10.56 %, 0.84 %, 30.06
%, 10.71 %, 30.02 %, 1.86 %, 1.055 % respectively
towards ganoderic acid B production by Ganoderma
lucidum under submerged fermentation. Fermentation
was carried out under shake flask condition with
10% v/v seed culture and incubated for 4 days under
shaking conditions and under static condition for 8 d
at temperature 27 0C. Among nine nutrient parameters
urea and NaCl contributed maximum while corn flour
and soya bean contributed moderately and MnSO4.H2O,
maltose, glucose, biotin and elicitors had little impact on
production of ganoderic acid B by Ganoderma lucidum.
